Mass Spectrometry (Imaging) at MR Core Facility
MR Core Facility offers access to gas chromatography (GC) mass spectrometry (MS) and matrix assisted laser desorption ionization (MALDI) - time of flight (TOF) MS equipment located in our labs. Our MALDI-TOF instrument is a Bruker rapifleX™ MALDI Tissuetyper™ specifically designed for high-speed mass spectrometry imaging (MSI) but can also be used for classical “non-imaging” MALDI-TOF mass spectrometry, including tandem MS, protein identification, drug screening, and oligonucleotides sequencing.

- 2 GC-MS systems from Agilent (7890A GC system coupled with a 5975 MS detector)
- Both equipped with electron ionization sources with the option to switch to chemical ionization (methane) sources
- Multiple columns for fatty acid, lipid, bile acid, amino acid analysis available
- MALDI-TOF/TOF mass spectrometer, Bruker rapifleX™ MALDI Tissuetyper™
- 10 kHz laser allowing fast MS image acquisition with up to 40 pixel/second
- High-speed non-imaging MS as fast as ~ 0.3 s/sample possible
- Robust, reliable, and easy operation
- HTX M5 Sprayer – MALDI matrix sprayer required for MSI applications
- High-velocity XY stage

Mass Spectrometry Imaging (MSI)
MSI is based on acquiring mass spectra pixel by pixel on tissue sections, which allows the spatial distributions of hundreds of different masses of for example metabolites or phospholipids to be captured at a time. All types of tissue sections and cell culture samples can be measured, from human heterogeneous tumor samples to plant tissue to organoid cultures. The rapifleX instrument at our facility has a supported maximal spatial resolution of 20 µm, but we could successfully image at even higher resolutions of 10 µm and 5 µm. This technique is nondestructive for most of the tissue section, meaning that the remaining tissue can be subjected to a broad variety of histology staining afterwards, allowing for a direct comparison between MSI and histology.
Special application MSI: Multiplexed MALDI-IHC
Multiplexed MALDI-IHC is the combination of immune histochemistry (IHC) with MALDI MSI for detection of mass-tag labeled anti-bodies. This technology allows extreme high-plexity by simultaneous staining and detection of hundreds of targets on the same tissue section. Multiplexed MALDI-IHC can be combined with conventional untargeted MSI (for example for metabolite and phospholipid imaging) and conventional microscopy-based IHC on the same tissue section for maximal utilization. We have required equipment and established protocols to perform the Miralys highplex assay from AmberGen readily available at MR Core Facility.
Peptide Mass Fingerprinting and other MS-based Proteomics Applications
MR Core Facility provides a joint service together with the Proteomics and Modomics Experimental Core Facility (PROMEC) for conventional MALDI-MS-based proteomics where liquid-chromatography is not required. Sample preparation can be performed at PROMEC while the MS analysis can be performed on our MALDI-TOF instrument. Please contact us for more information.
