Zeiss 700 Confocal Microscope

Zeiss 700 Confocal Microscope

– Upright confocal microscope

The LSM 700 microscope is equipped for standard confocal imaging of biological specimens and animals, as well as for materials examination. The following microscopy and contrasting techniques are possible:

  1. Time lapse imaging
  2. Emmision fingerprinting
  3. Differential interference contrast (DIC) in combination with fluorescence
  4. Polarized light microscopy
  5. Epifluorescence and confocal imaging of thick specimens (animals)

The microscope

The Axio Imager.Z1 is an upright, motorized microscope with a detachable scanning stage and touch pad.

Focus drives

  • Motorized stage (z-step size 30 nm)

Scanners

  • Galvo scanning, up to 5 frames /sec at 512 x 512 pixels image format can be achieved

Objectives

Magnification NA Type Immersion Coverslip WD (mm)
10x 0.3 EC Plan Neofluar Air -/ 0.17 5.2
20x 0.8 Plan Apochromat Air 0.17 0.55
40x 1.0 Plan Apochromat Water (dipping) - 2.5
40x 1.3 C-Apochromat Water Oil 0.21
63x 1.2 C-Apochromat Water 0.14 - 0.17 (Corr ring) 0.28
  • DIC prisms exists for the following objectives: 20x/0.8, 40x/1.0, 40x/1.3 and 63x/1.2

Condenser lens

Motorized condenser, NA 0.90 air, working distance 1.2 mm

Lasers

  • Solid-state lasers 405 nm (5 mW), 488 nm (10 mW), 555 nm (10 mW) and 639 nm (5 mW)
  • An acousto-optical tunable filter (AOTF) controls the intensity of the visible excitation laser light
  • One main beamsplitter is used for all laser lines to separate excitation light from the emitted photons

Detectors

Two internal PMT detectors (with pinhole) in reflection mode with tunable emission bands in the range 400 – 700 nm.

One external PMT (without pinhole) in transmission mode.

Light for epi-fluorescence and transmitted light

  • External HXP 120W, fiber optics and bulb lifetime 2000h
  • Halogen lamp: 12V – 100W

Accessories

  • The scanning stage including condenser optics can be removed. A custom made stage (no XY-control and condenser optics) can be directly connected to the microscope stand, utilizing the z-focusing drive.
