Motivation

To study the relationship between morphology and function in ‘cell types’, we will update our targeting of genetic cell types47 to incorporate single-cell technologies (scEDGE) and use it to target genetic cell types and test their functions in circuit development and plasticity.

Aim

4.1 To identify and manipulate genetically-defined circuit components through the use of scEDGE

Implementation

Using scATACseq to assign ChIPseq marks to particular genetic cell types, we shall engineer scEDGE-AAVs that specifically target genetically-defined cell types in specific cortical regions (WPs1-3) and by doing so determine functions of these cell types in neural circuit development (WP3A) and plasticity.
